Este filme conta a história de 5 detectives, Fred, Daphne, Shaggy, Velma e Scooby Doo, que põem fim ao grupo por causa de uma incompatibilidade de egos.

Passado algum tempo todos são convidados individualmente para fazerem uma viagem até Spooky Island, para desvendarem uma série de mistérios que têm acontecido aos visitantes da ilha.

Mas para conseguirem resolver o caso em conjunto eles terão de colocar os seus desentendimentos de lado e fazer o que cada um sabe fazer de melhor.
